Article Abstract:
The expression vector lambdaYES was used to construct a copyDNA library for Phanerochaete chrysosporium . This vector has a lacZ promoter and GAL1 promoter for the expression of both Escherichia coli and Saccharomyces cerevisiae, respectively. These characteristics enabled the complementation of auxotrophs for E. coli and yeast and allowed for the cloning of the leu2 gene as well as other cDNAs involved in biosynthetic pathways. A potential application for this would be in the transformation of P. chrysosporium and other wood-degrading fungi.

Article Abstract:
A new robust expression system for Bacillus subtilis conditional complementation is presented, contained on plasmid pSWEET, which is a derivative of pDG364 (3). Testing involved comparing the system to the isopropyl-beta-D-thiogalactopyranoside-induced spac system, which determined that the xylose-dependent expression derived from pSWEET had a greater modulated expression capacity, a higher induction/repression ratio, and lower expression levels if no inducer is present.

Article Abstract:
Research was conducted to study several food-grade bacterial strains of the genus Bacillus subtilis for extracellular phytase production. Strains were evaluated for phytase production in Luria broth while bacteria prepared on wheat bran extract were gathered by centrifugation. Moreover, the reaction pH was represented as a function of relative activity. Results showed that all cloned and sequenced microbial phytases were homologous to each other.
