Cloning, nucleotide sequence, and transcriptional analysis of the Pediococcus acidilactici L-(+)-lactate dehydrogenase gene

Article Abstract:

Complementation of an Escherichia coli lactate dehydrogenase-pyruvate formate lyase double mutant helps clone recombinant plasmid having the Pediococcus acidilactici L-(+)-lactate dehydrogenase gene (ldhL). A protein of 323 amino acid present in the ldhL gene exhibits similarity with L-(+)-lactate dehydrogenases in Lactobacillus plantarum. Primer extension analysis helps define ldhL transcription start points in P. acidilactici. The promoter sequence of P. acidilactici is significantly related to the consensus sequence of vegetative promoters for both gram-positive bacteria and Escherichia coli. Experiments reveal the expression of ldhL gene as a monocistronic transcript in P. acidilactici, and growth rate regulates this transcription.

Cloning, expression and nucleotide sequence of genes involved in production of pediocin PA-1, a bacteriocin from Pediococcus acidilactici PAC1.0

Article Abstract:

The presence of the plasmid pSRQ11 is required for the production of pediocin PA-1. Sequencing of the plasmid revealed that it contained four genes coding for proteins which were involved in pediocin production. The pedA gene encoded a precursor sequence for pediocin PA-1. The pedD gene coded for a protein which is required for pediocin production in Escherichia coli and is similar to ATP-dependent transport proteins, suggesting a role in translocation. No functions could be assigned to pedB and ped C. The use of E. coli vectors in cloning the plasmid showed that the pediocin is produced and may be secreted by E. coli.

Expression of a Lactococcus lactis phage resistance mechanism by Streptococcus thermophilus

Article Abstract:

Greater phage resistance was observed in Streptococcus thermophilus for the first time. The 7.8-kb lactococcul plasmid pSRQ700 encoding the LlaII modification-restriction system is unable to replicate in Streptococcus thermophilus. The lytic or temperate S. thermophilus have small isomeric heads and noncontractile tails. Suitable introduction of LlaII gene into S. thermophilus may confer effective phage resistance and stability without selective pressure at 37 and 42 degree centigrade.
