Article Abstract:
The enhancer trap approach was adopted in searching for genes which play important roles during oogenesis. A Drosophila melanogaster line whose nurse cell and oocyte nuclei expressed beta-galactosidase was selected and chromosome walking was used to clone DNA flanking the P(lac, ry+)A transposon. Within a transcription unit cluster expressed in the ovary, a transcription unit encoding a putative ubiquitin carboxyl terminal hydrolase was identified. After confirming the presence of the appropriate enzyme activity in a fusion protein, the gene was named ubiquitin carboxyl terminal hydrolase uch-D.

Article Abstract:
The structure of actin-containing cortical cytoskeleton of developing oocytes of the silkmoths Hyalophora cecropia and Antheraea polyphemus was investigated. In addition, the process of mRNA localization in the oocytes and the role of RNA polymerase II (pol II) in determining actin filament distribution pattern were sttudied. Immunohistochemical analysis coupled with confocal laser scanning microscopy were used in the study. The results indicate that the distribution of both mRNA and pol II in the cortex depends on the integrity of the actin scaffolding in the cortex.
