Article Abstract:
The crystal structure of clathrin has been the subject of many studies on proteins. While attempts to crystallize the molecule have not been successful, recent gains in determining the crystal structure of recombinant terminal domain and linker, and production of recombinant hubs may produce a full picture of the clathrin molecule. In addition, a map of triskelion packing in the clathrin cage in conjunction with a representation of the folding pattern of heavy chain and light chain subunits in the triskelion will offer a framework for understanding clathrin assembly properties.

A compendium of chemical-genetic interaction profiles was generated by testing the collection of viable yeast haploid deletion mutants for hypersensitivity to 82 compounds and natural product extracts. Hierarchical clustering analysis and probabilistic sparse matrix factorization reveals that tamoxifen, a breast cancer therapeutic, disrupts calcium homeostasis and identifies phosphatidylserine as a target for papuamide B, a cytotoxic lipopeptide with antifungal and anti-human immunodeficiency virus activity.

This review elucidates the function of the transmembrane receptor cytoplasmic domain (tail)-binding proteins such as Adaptor Protein, Golgi localized gama-adaptin, and other tail binding proteins in sorting lysosomal enzyme receptors in mammalian and yeast cells.
