Article Abstract:
The Sry gene is found at higher levels in the adult mouses testis than in the genital ridge which is at a critical site of action. RNAase protection assays and reverse transcription polymerase chain reactions reiterate that these unusual RNA molecules are the most abundant transcript in testis. Oligonucleotide hybridization and RNAase H digestion confirm that these Sry RNA molecules are circular. The Sry RNA molecules are circular due to the unique genomic structure encircling the Sry locus in the mouse.

Research has provided a molecular mechanism that determines gender in mammalian embryos. The process involves several stages, including general transcription factors, specific molecular promoters of testis development and ongoing regulatory loops such as cell-cell communication to combat ovary development. Further research is expected to uncover processes linking steroidogenic and supporting cell lineages.

Mutagenesis of the endogenous mouse Mis gene promoter is discussed with consideration of in vivo definition of pathways of vertebrate sexual development. SOX9 appears to be essential in initiation of Mis transcription. SF1 seems to serve as a quantitative regulator of Mis transcription levels. The Mis gets transcription inputs that vary with species but give the same functional readout.
