The putA and putP genes of Pseudomonas putida KT2440 have separate promoters and are transcribed divergently. The PutP protein takes up the amino acid proline, which is the bacterium's sole source of C and N, and the PutA protein converts proline to glutamate.

The Pseudomonas putida KT2440 TOL upper pathway is repressed under nonlimiting conditions in cells growing in chemostat with succinate as a carbon source. A demonstration that the ptsN gene product [IIA.sup.Ntr] participates in this repression is presented.

The expression of the Pseudomonas putida toluene and m- and p-xylene degradation (TOL) upper pathway is low under phosphate and sulfate limiting conditions. TOL expression is high under oxygen, carbon or energy limiting conditions and intermediary under ammonia limiting conditions. Succinate is the sole carbon and energy source under all conditions. The catabolic repression of TOL is due to the high energy status of the cell. The suppression is associated with low levels of the upper pathway promoter mRNA.
