Article Abstract:
The genomic organization of the nisin biosynthetic genes in Lactococcus lactis 6F3 was elucidated. Analysis was conducted on the adjacent regions to the nisin structural gene, nisA. Three L. lactis genes were isolated and characterized. Based on homologies to other lantibiotic structural genes, the isolated genes were designated nisB, nisT and nisC. The ordering of the genes relative to the nisA structural gene is nisA, nisB, nisT, nisC. Western immunoblot analysis revealed a molecular size of 115-kDa for the NisB protein, which was localized to the vesicle fraction of the cell membrane. This indicates that nisin biosynthesis occurs within the cell membrane.

Experimental studies on Lactococcus lactis 6F3 reveal the presence of three additional open reading frames nisG, nisE and nisF in the nisin gene cluster. Mutants with mutations in these genes exhibit higher sensitivity to nisin, revealing the involvement of these genes in immunity to nisin. Significant homology is observed between proteins, NisF and NisE, and members of the family of ATP-binding cassette transporters.

Article Abstract:
Identification and genetic analysis were done on reading frames upstream of the Bacillus subtilis biosynthesis spaS. The reading frames spaB, spaC and spaT showed similar homologies to open reading frames identified near the structural gene of the lantibiotic epidermin. However, gene deletion studies showed that only the reading frames spaB and spaC were essential for subtilin biosynthesis.
