Biochemical and molecular characterization of the polyhydroxybutyrate depolymerase of Comamonas acidovorans YM1609, isolated from freshwater

Article Abstract:

A research was conducted to study the purification of the extracellular polyhydroxybutyrate (PHB) depolymerase from the culture supernatant of Comamonas acidovorans and to examine its biochemical and kinetic properties. The function of the putative substrate-binding domain was determined through a fusion protein with glutathione S-transferase formed in Escherichia coli. Results indicated that the PHB depolymerase was composed of discrete domains connected by a linker region.

Poly(aspartic acid) degradation by a Sphingomonas sp isolated from freshwater

Article Abstract:

Research was conducted to examine the biodegradability of thermally synthesized poly(aspartic acid) (PAA) polymers in natural river water. A PAA-degrading bacterium was isolated from freshwater and its biodegradabilities were calculated by biochemical oxygen demand assay and by gel permeation chromatography analysis. Results indicate that degradation of PAA by strain KT-1 has similar characteristics with the PEG degradation by Sphingomonas macrogoltabidus.

Genetic analysis of comamonas acidovorans polyhydroxyalkanoate synthase and factors affecting the incorporation of 4-hydroxybutyrate monomer

Article Abstract:

Research was conducted to examine the cloning and characterization of the polyhydroxyalkanoate (PHA) synthase gene of Comamonas acidovorans, DS-17. Substrate specificity of C acidovorans was analyzed using as heterologous hybridization probe the synthase gene of Alcaligenes eutrophus. Results indicate that the monomer-supplying pathway plays a significant role in determining the type of PHA that can be produced.
