Binding of a novel SMG-1-Upf1-eRF1-eRF3 complex (SURF) to the exon junction complex triggers Upf1 phosphorylation and nonsense-mediated mRNA decay
Article Abstract:
The SMG-1 immunoprecipitates (SMG-1-IP) for the presence of proteins that associates with MRNP were analyzed, to explore the possibility that the phosphorylation of Upf1 by SMG-1 occurs on mRNAs. The findings show that the SMG-1-mediated phosphorylation of Upf1 occurs on the association of SURF with EJC (exon junction complex), which provides the link between the EJC and recognition of PTCs (premature termination codons) and triggers NMD (nonsense-mediated mRNA decay).

The different classes of mammalian poly(A) nucleases are tested. A new insight into a mechanism is provided by which interplay between mRNA turnover and translation determines the lifespan of a mCRD-containing mRNA in the cytoplasm.

The article describes that let-7 miRNPs containing Argo -naute and GW182 impair the synergistic enhancement of translation by 5'cap and 3'-poly(A) tail, which results in translational repression.
