Basin-wide analysis of the dynamics of fecal contamination and fecal source identification in Tillamook Bay, Oregon

Article Abstract:

A study was conducted to examine spatial and temporal dynamics in source-specific Bacteriodales 16S rRNA genetic marker data across a watershed and compare these dynamics to fecal indicator counts. Inconsistencies in Escherichia coli and host-specific marker trends suggested the factors that control the quantity of fecal indicators in water column are different than factors that influence presence of Bacteroidales markers at specific times of the year.

Microplate subtractive hybridization to enrich for bacteroidales genetic markers for fecal source identification

Article Abstract:

Subtractive hybridization was used to enrich for target host-specific fecal Bacteroidales rRNA gene fragments that were different from those very closely related reference host source. The results demonstrate the method's ability to enrich for variable regions of the 16S rRNA gene and that this method could readily be adapted for gene targets.

Rapid estimation of numbers of fecal Bacteroidetes by use of a quantitative PCR assay for 16S rRNA genes

Article Abstract:

The quantitative Taq nuclease assay (TNA) for general fecal pollution using a Bacteroidetes 16S rRNA marker is reported. The sensitivity and correlation with standard fecal indicators provide experimental evidence for application of the assay in monitoring fecal pollution.
