Analysis of the transcriptional program induced by Raf in epithelial cells

Article Abstract:

To study the contribution of MAP kinase-mediated transcriptional changes to the transformed phenotype an inducible form of Raf was used to study early changes in the transcription of some 6000 genes after activation of the kinase in a normal human breast epithelial cell line. Autocrine activation of the EGF receptor was seen to control ability of RAf activation to protect the cells from apotosis brought on by detachment of cells from extracellular matrix.

BH3-only proteins that bind pro-survival Bcl-2 family members fail to induce apoptosis in the absence of Bax and Bak

Article Abstract:

BH3-only proteins that bind pro-survival Bcl-2 family members have been found to not induce apoptosis without Bak and Bax. It appears that neither activation of BH3-only proteins nor suppression of pro-survival Bcl-2 proteins can kill cells without them. Oncogenic transformation, it appears, is supressed by the function of either Bax or Bak.

The serine/threonine kinase Pim-2 is a transcriptionally regulated apoptotic inhibitor

Article Abstract:

Research has been conducted on serine/threonine kinase Pim-2. The authors reveal that Pim-2 is a direct link between growth factor-induced transcription and kinase-dependent pathway which promotes cell-autonomous survival.
