Immunoprecipitation and mass spectrometry have been used to identify BRCA1-associated proteins. It was found that BASC (BRCA1-associated genome surveillance complex), a super complex of BRCA1-associated proteins, is involved in recognition/repair of DNA structures that are defective.

Article Abstract:
Functional interactions between BRCA1 and the checkpoint kinase ATR under genotoxic stress have been studied. ATR, related to ataxia-telangiectasia-mutation, was found to respond to DNA damage and replication blocks by forming distinct nuclear foci at the sites of stalled replication forks. Based on this and other findings, it was concluded that ATR and BRCA1 are parts of the same genotoxic stress-responsive pathway. ATR directly phosphorylates BRCA1 as a response to damaged DNA or stalled DNA replication.

Article Abstract:
Results show that DNA damage by cyclin-dependent kinase leads to phosphorylation of the anaphase inhibitor Pds1 at multiple sites, which stabilizes Pds1. Data further indicate that mutations in the Pds1 phosphorylation sites abolish its DNA dmage-inducible phosphorylation and kinase functions.
