Assessment of (3H)thymidine incorporation into DNA as a method to determine bacterial productivity in stream bed sediments

Article Abstract:

The applicability of the (3H)thymidine incorporation method in measuring bacterial productivity in stream bed sediments was studied. The results showed that this procedure could yield reasonable values. However, some limitations exist. Firstly, some aerobic and anaerobic bacteria failed to incorporate thymidine into their DNA. Secondly, the determination of isotopic dilution requires a better understanding of the biochemical pathways for thymidine. And lastly, a model for the conversion of DNA incorporation rates into cell production is lacking.

Evaluation and simplification of the assimilable organic carbon nutrient bioassay for bacterial growth in drinking water

Article Abstract:

A modified assimilable organic carbon (AOC) bioassay was proposed. All aspects of the AOC bioassay, including inoculum, incubation water, bioassay vessels and other glassware and enumeration technique, were evaluated. Both bioassay organisms, Pseudomonas fluorescens P-17 and Spirillum sp. strain NOX, are available from the American Type Culture Collection and can be easily grown and maintained. Quality control includes an AOC blank and checks for organic carbon limitation and inhibition of the bioassay organisms.

Development of a rapid assimilable organic carbon method for water

Article Abstract:

Arapid method for determination of assimilable organic carbon (AOC) was proposed. The time needed to perform the assay is reduced by increasing the incubation temperature, increasing the inoculum density and using the ATP luciferin-luciferase method to quickly enumerate the test organisms. The results show a good correlation between the rapid AOC technique and the plate count procedure.
