Assay of poly(3-hydroxybutyrate) deplolymerase activity and product determination

Article Abstract:

Two methods for accurate poly(3-hydroxybutyrate) (PHB) depolymerase activity determination and quantitative and qualitative hydrolysis product determination are described. The first method is universally suitable for hydrolysis of any type of polyhydroxyalkanoate or other molecules with hydrolyzable ester bonds, allows the determination of hydrolysis rates of as low as 1 nmol/min, and has a dynamic capacity of at least 6 orders of magnitude. The second method allows the separation and quantification of 3HB and 3HB oligomers up to the octamer.

Altered specificity of lactococcal proteinase PI (Lactocepin I) in humectant systems reflecting the water activity and salt content of cheddar cheese

Article Abstract:

The specificity of hydrolysis of alpha-s1-, beta-, and kappa-caseins by the cell envelope-associated proteinase lactocepin from Lactococus lactis subspecie lactis BN1 was examined at water activities [a(sub w)] and salt concentrations reflecting those in cheddar cheese by using various humectant systems. Findings showed that the specificity of lactocepin was dependent on a (sub w) and medium hydrophobicity. It was clear that the characteristic differences of the lactocepin types were not fully reflected in the cheese environment.

Applications of a rapid endospore viability assay for monitoring UV inactivation and characterizing arctic ice cores

Article Abstract:

The endospore viability assay (EVA) against parallel experiments was developed and tested by using phase-contrast microscopy and traditional heterotrophic plate counts. It was developed as a rapid alternative to culture based assays for quantifying the efficiencies of sterilization, bioburden reduction and decontamination operations and the percentages of germinable endospores embedded in arctic ice cores.
