Argonaute2 cleaves the anti-guide strand of siRNA during RISC activation

Article Abstract:

The mRNA-cleavage step of RNA interference is mediated by an endonucleus, argonaute2 (Ago2) within the RNA-induced silencing complex (RISC). The anti-guide strand behaves as a RISC substrate and cleaved by Ago2, which is important for the removal of the anti-guide strand from Ago2 protein and activation of RISC.

Neutrophil elastase cleaves PML-RARalpha and is important for the development of acute promyelocytic leukemia in mice

Article Abstract:

Research demonstrates that neutrophil elastase mediates the fusion protein PML-RARalpha initiation of acute promyelocytic leukemia. Results show that the neutrophil elastase is produced in promyelocytes. Data indicate that hematopoietic proteases may have a role in leukemia development.

RNAi: RISC gets loaded

Article Abstract:

Simplified views of siRNA-programmed RISC activation are explored. A heterotrimeric complex from humans is defined that appears to execute dsRNA loading, strand selection, and target mRNA cleavage activities.
