Analysis of sequences flanking the vap regions of Dichelobacter nodosus: evidence for multiple integration events, a killer system, and a new genetic element

Article Abstract:

DNA sequences flanking the virulence-associated protein (vap) regions 1,2 and 3 were analysed using the Southern blotting and DNA sequencing. These vap regions of the Dichelobacter nodosus are found in 98% of virulent strains tested which suggest that they may have a role in virulence. They appear to have arisen by the integration of a genetic element into a tRNA gene. A new genetic element integrated into the Dichelobacter nodosus chromosome of vap region 3 was identified as either an integrated bacteriophage or conjugative transposon.

A gene region in Dichelobacter nodosus encoding a lipopolysaccharide epitope

Article Abstract:

A Dichelobacter nodosus locus that modified an Escherichia coli K12 lipopolysaccharide (LPS) was cloned and sequenced for examination of its genetic structure in an effort to identify any changes that could cause footrot in sheep. Changes were identified in the inner-core region of the heptose molecules found in the LPS. Previous research suggests that the outer membrane of D. nodosus is affected by the structure of the LPS, and allows the gram negative bacteria to affect and by carried by the sheep.

Organization of ribosomal RNA genes from the footrot pathogen Dichelobacter nodosus

Article Abstract:

Southern hybridization, sequence analysis and primer extension analysis were used to locate and determine the characteristics of the three rrn loci within the genome of Dichelobacter nodosus. These loci were found to be composed of 16S, 23S and 5S rRNA genes. Putative genes encoding tRNA(super Ile) and tRNA(super Ala) were found within the 16S-23S spacer region. Possible open reading frames were discovered within the regions adjacent to the rrn loci.
