The sequence analysis of the Tox5 promoter regions from geographically significant strains of Gibberalla pulicaris demonstrate the existence of Tox5-1 and Tox5-2. Strains producing high levels of trichothecenes carry a 42-nucleotide (nt) repeat sequence (Tox5-1), while those with low levels carry a single copy of the sequence (Tox5-2). Genetically crossing high and low-level trichothecene producers gives a cosegreggation of higher-level trichothecene production with Tox5-1.

The acetyltransferase gene (Tri3) of Fusarium sporotrichioides is related to the three genes of the trichothecene biosynthetic pathway. Tri3 has four introns. The Tri3 open reading frame encodes a protein with a molecular weight of 57,418 Da. Tri3 encodes the 15-O-acetyltransferase, which converts 15-decalonectrin to calonectrin. The disruption of the gene produces an excess of the deacetylated calonectrins and the blockage of 15-O-acetylation.

Article Abstract:
Research was performed to determine the characterization of TRI11 gene from the trichothecene gene cluster in Fusarium sporotrichioides. The nucleotide sequence of TRI11 predicts a polypeptide of 492 residues with considerable likeness to members of the cytochrome P-450 superfamily. Evidence suggests that disruption of TRI11 leads to an altered trichothecene production phenotype characterized by the accumulation of isotrichodermin.
