Article Abstract:
An effective approach for the detection of coated DNA in natural seawater is discussed. Eutrophic seawater samples were derived from Tokyo Bay and subjected to enzymatic differentiation of free and coated DNA materials, DNA size fractionation and immunochemical-potentiometric DNA quantification. DNA molcular size was also estimated. The dominance of coated DNA in eutrophic seawater was established.

Article Abstract:
The optimization of fluorescence in situ hybridization (FISH) procedure using rRNA-targeted oligonucleotide probes and flow cytometry for the detection of Escherichia coli in seawater supplies is reported. The optimized protocols were applied to E. coli-spiked seawater samples and had a good recovery rate of the spiked E. coli cells.

High incubation temperatures and a lysis buffer with high concentrations of Tris, EDTA, NaCl, SDS, and proteinase K are required to extract Brucella DNA from bovine milk samples. PCR can then be used to amplify the DNA.
