Exploring the transcription-chromatin interface

Article Abstract:

The transcription-chromatin interface is discussed in this meeting-review article covering a workshop on 'Integration of Transcriptional Regulation and Chromatin Structure.' It was hosted by the Fundacion Juan March in Madrid, Spain. Sessions included those on functions of specialized core promoter structures, promoter/enhancer -binding factors, locus control regions, and boundary elements. Research has shown that factors can be activators in one situation and repressors in another. Topics include ATP-dependent chromatin remodeling complexes, chromatin-modification complexes, other coregulators, and gatekeepers.

Enhancer-promoter specificity mediated by DPE or TATA core promoter motifs

Article Abstract:

Recent study has shown the existence of transcriptional enhancers that are specific for promoters that contain either TATA box or downstream promoter element (DPE) elements. The core promoter is seen to mediate initiation of transcription and also to act as a regulatory element. The ability of enhancers to activate transcription in vivo from core promoters having either DPE or TATA box motifs was studied to look into the basis for enhancer-promoter specificity.

ACF consists of two subunits, Acf1 and ISWI, that function cooperatively in the ATP-dependent catalysis of chromatin assembly

Article Abstract:

ATP-utilizing chromatin assembly and remodeling factor (ACF) is made up of Acf1 and ISWI, two subgroups that cooperate in ATP-dependent catalysis of chromatin assembly. Drosophila acf1 cDNA has been isolated. It encodes the p170 and p185 forms of the Acf1 protein in ACF. Acf1 is a novel protein containing two PHD fingers a bromodomain and two conserved regions that are new. Human WSTF, encoded by one of multiple genes deleted in those with Williams syndrome, is the only known mammal protein with all of the conserved motifs in Acf1. The activity of ISWI alone is about 3% of that of ACF.
