A tyrosine-based signal targets H/K-ATPase to a regulated compartment and is required for the cessation of gastric acid secretion

Article Abstract:

Researchers have investigated the role of a tyrosine-based sequence motif present in the cytoplasmic tail of the beta subunit of the H/K-ATPase in the targeting of this protein to a regulated compartment in vivo. This work involved mutagenizing the Tyr residue to Ala and producing transgenic mice expressing either the wild type or the mutagenized form of the H/K-ATPase beta subunit. It was established that the H/K-ATPase beta subunit tyrosine-based motif is required to prompt the removal of the H/K-ATPase from the apical membrane of parietal cells. The motif is also need to ensure that the H/K-ATPase is returned to the TVE compartment to stop the process of acid secretion.

Lymphoid potential, probed before circulation in mouse, is restricted to caudal intraembryonic splanchnopleura

Article Abstract:

The intraembryonic precursor source in the paraaortic splanchnopleura (P-Sp) and aorta-gonad-mesonephros is investigated using a culture system of isolated mouse Sp. Dissection of Sp is made prior to circulation. Hemopoietic intraembryonic precursors are observed even at the presomite stages or at 7.5-8 days postcoitum in yolk sac (YS) Sp. Comparison of Sp and YS precursors from one embryo shows larger differentiation ability and cultural survival for Sp precursors. YS has no capacity for a lymphoid progeny and has insufficient precursors for mixed myeloid colonies.

Globin gene expression is reprogrammed in chimeras generated by injecting adult hematopoietic stem cells into mouse blastocysts

Article Abstract:

The differentiation capacity of adult hematopoietic stem cells (HSC) is investigated by injecting HSCs from adult mice into blastocysts. Results reveal that adult HSCs survive in mouse embryos, further, the erythroid line synthesized fetal globin proteins. Conversely, embryonic erythroid progenies synthesize adult globin proteins when injected to adult mice. HSCs from adult mice exhibit gene expression plasticity depending on the environment.
