SUR-8, which is rich in leucine, enhances mitogen-activated protein (MAP) kinase (MAPK) activation according to this research communication article. It also forms a complex with Raf and Ras. Caenorhabditis elegans sur-8 encodes a regulator of Ras signaling in the positive sense. The mechanism through which the human sur-8 homolog can regulate Ras-MAP kinase signaling positively in mammalian cells has been investigated. The effect of expressing human sur-8 on the activity of ERK, a MAPK family member, in mammalian cells has been tested.

A probable G-protein signaling network mediated by one of the regulators of G protein signaling, or RGS proteins, is discussed. Mediation of a G-protein network in Caenorhabditis elegans by the EAT-16 RGS protein and G-protein cascade models are covered.

The in vivo substrate selectivity of Ptp3 toward the Fus3 mitogen-activated protein (MAP) kinase is discussed. The Saccharomyces cerevisiae mating pheromone response is a well-characterized MAP kinase (MAPK) signaling pathway for forming of diploid cells from haploid cells. The molecular mechanisms responsible for Pto3 substrate preference in vivo have been investigated. Substrate selectivity is determined by the amino-terminal noncatalytic domains.
