Article Abstract:
Examination of the electrotransformation of plasmid DNA into Streptomyces rimosus R6 reveals that buffer composition, electric field strength and pretreatment of mycelia significantly influence the transformation. This simple, fast electroporation method generates transformant colonies in 48 to 72 h. Experimental studies reveal that diversity of Streptomycetes is responsible for the requirement of different optimizations for different strain.

Article Abstract:
The growth and chitinolytic activity of Streptomyces lividans containing t he cloned exochitinase gene in a soil microcosm system are studied by in situ hybridization and microscopic analyses. Results show that the propagation rate of S. lividans is higher in soil microcosms amended with fungal mycelia than in those containing crab chitin. The data also indicate the presence of a novel chitin-binding protein that lacks chitinolytic activity.

The bacterium Streptomyces rimosus MA187 can demethylase the immunosuppressant FK-506 at the 31st carbon by using an enzyme that has been purified. This enzyme, 31-O-FK-506 demethylase resembles cytochrome P-450 and is most active at a pH of 7.4 and a temperature of 34 degrees Celsius. The enzyme can be used to generate a significant immunosuppressant and metabolic product of FK-506.
