A new approach to utilize PCR-single-strand-conformation polymorphism for 16S rRNA gene-based microbial community analysis

Article Abstract:

A study was conducted to characterize a novel technique for using polymerase chain reaction (PCR)-single-strand-conformation polymorphism in 16 rRNA gene-based microbial community analyses. Cells from pure cultures harvested to late logarithmic growth phase were pelleted in tubes by centrifugation while the phosphorylated strand was removed by lambda exonuclease digestion to determine single-stranded DNA from PCR products. Single bands identified in polyacrylamide gels were then cut out for further analysis.

Article Abstract:

Research has been conducted on the Norwalk-like viruses and hepatitis A in shellfish. The development of the extraction-and-detection procedure that has led to the RT-PCR amplification of limited quantities of the hepatitis A and Norwalk-like viruses in shellfish extracts is reported.
