Characterization of IS900 loci in Mycobacterium avium subsp. paratuberculosis and development of multiplex PCR typing

Article Abstract:

Researchers describe 14 of the 18 copies of IS900 insertions in the genome of Mycobacterium avium subsp. paratuberculosis. The DNA flanking the insertions was analyzed and homologues were found in Mycobacterium tuberculosis and Mycobacterium avium subsp. avium.

IS900 targets translation initiation signals in Mycobacterium avium subsp. paratuberculosis to facilitate expression of its hed gene

Article Abstract:

The atypical insertion sequence (IS) IS900 present in Mycobacterium avium subsp. paratuberculosis genome targets translation initiation signals to facilitate expression of the new gene designated as hed (host-expression dependent). A promoter, ribosome binding site (RBS) and termination codon are all needed to be encoded by sequences that flank IS900. The element IS900 inserts between the RBS and start codon of a putative gene sequence hence aligning the hed initiation codon adjacent to a functional RBS.

A low G+C content genetic island in Mycobacterium avium subsp. paratuberculosis and M. avium subsp. silvaticum with homologous genes in Mycobacterium tuberculosis

Article Abstract:

The genetic differences between the enteric pathogens, Mycobacterium avium paratuberculosis and Mycobacterium avium silvaticum, were studied using representational difference analysis. The principal distinction between the genomes of M. avium complex strains was perceived to be the presence of transposable insertion elements including IS1245, IS900 and IS901/2. The nucleotide sequence of the pathogens' gene specific (GS) regions showed five open reading frames in a distinct region. The G+C content and the relation with a potential mobilization factor were features of the GS element.
