Article Abstract:
A library of vectors expressing the majority of cloned human miRNA was developed and corresponding DNA barcode arrays is created to perform genetic screens for novel functions of miRNAs. The results provide evidence that these miRANs are potential novel oncogenes participating in the development of human testicular germ cell tumors by numbering the p53 pathway, thus allowing tumorigenic growth in the presence of wild-type p53.

RNA interference libraries targeting large proportions of the human genome are used to uncover two tumor suppressor genes, REST and PITX1. While REST is a transcriptional repressor that silences neuron-specific gene expression, PITX1 is a homeodomain transcription factor that promotes the expression of a negative regulator of Ras.

An RNA-interference library for knockdown constructs that conform human primary cells in the absence of ectopically introduced oncogenic RAS gene was screened to uncover novel tumor-suppressor genes. The identification of PITX1, whose inhibition induces the RAS pathway and tumorigenicity, is reported.
